    摘要: 聚多巴胺是具有前景的材料,且已被應用至廣泛領域。然而,目前研究發展對於聚多巴胺性質的瞭解還是有限,這驅使了許多學者試圖探討其未知的特性,以利於新穎應用開發。在此研究中,我們主要探討鹼催化法形成之聚多巴胺顆粒之性質,以拓展其於生物醫學領域之應用。因此,本論文分三個部分進行,包括(1)以鹼催化法製備聚多巴胺顆粒與其於抗癌治療之應用、(2)以鹼催化共析出法製備二氧化矽?聚多巴胺複合物,並進行定性分析、(3)探討二氧化矽?聚多巴胺複合物用於提升骨生成性之鈦表面處理之可行性。在第一部分,我們了解到溶液酸鹼度對於聚多巴胺顆粒形成之影響,可以用以作為非水溶性抗癌藥物之載體。第二部分,我們提出於弱鹼環境下,可利用共沉析法製備具階級結構之二氧化矽?聚多巴胺複合物,且可藉由調控合成溶液酸鹼度以控制複合物之型態,製備出絮凝型與顆粒型複合物。 第三部分,二氧化矽?聚多巴胺複合物可於合成時被塗覆於鈦金屬表面,且可藉由提升骨母細胞於鈦金屬之增生、分化以及礦化能力,以改善鈦金屬之骨生成性。
    Polydopamine (PDA) has emerged as a promising material and has been applied for a wide spectrum of applications. However, the full spectrum of PDA remains unclear, which prompts a great amounts of researches attempts to understand the properties of PDA for developing novel applications for different realm. In the present study, we focuses on physiochemical properties of PDA particles synthesized through based-catalyzed route for expanding the uses of PDA in biomedical applications. Thus, this study is divided into 3 parts, including (1) base-catalyzed formation of PDA particles and the application on anticancer therapy, (2) fabrication and characterization of silica/PDA hybrids synthesized through co-precipitation in basic solution, and (3) its application on the surface modification of titanium for improving the osteogenicity. In part 1, size-controllable PDA particles was fabricated by self-oxidation and assembly of DA in NaOH solution. The precursor’s pH value plays a crucial role on the particle size and yield of PDA. The PDA particles can serve as carriers for loading water-insoluble anticancer drug, camptothecin, with slow release behavior, and shows anticancer activity. For part 2, we proposed a co-precipitation of silica and PDA route for fabricating silica/PDA hybrids with hierarchical structure in an emulsion system under weakly basic condition. The results indicated that the morphology of silica/PDA can be controlled by adjusting solution pH to obtain flocculated form and particulate form of hybrids. In part 3, silica/PDA hybrids was successfully coated on titanium surface through the co-precipitation route, and the osteogenicity, including proliferation, differentiation and mineralization of osteoblasts cultured on titanium, was remarkably improved after silica/PDA coating.
