    Title: 荷葉類黃酮多重護肝及抑制肝癌之作用及機轉之研究

    Abstract: 荷?萃取物(Nelumbo nucifera leaves extract)具有豐富的?黃酮成分。過去幾?本實驗室已針對荷?萃取物(NLFE)以及?黃酮萃取物(NLFRE)進?研究,已指出荷?的粗萃物具有極佳的抗氧化能?,並可以抑制LDL的氧化;在動物實驗方面已經證實荷?萃取物能抑制動脈粥?硬化發生(附件一)、?低血脂及排除肝脂肪堆積(附件二);細胞方面?觀察到可有效抑制平?肌細胞的增生與移動(附件三),進一步也發現能抑制體脂肪的生成(附件四)。因此,我們將研究延緩各種病因引起之脂肪肝、護肝作用及抗肝癌作用之機轉,研究分為(1) NLFE延緩各種致病因子引起之脂肪肝護肝作用;(2)NLFE抑制肝癌化之作用及機轉;及(3)NLFRE對各種?同病因致肝損傷及肝癌化之保護作用機轉。第一?分析荷?萃取物(NLFE)對於高?+STZ(type 2 DM)、化學性(APAP)、細菌性(LPS),及酒?性肝病變之保肝作用;第二?進一步以動物模式探討NLFE抑制肝纖維化及肝癌之作用機轉;第三?以肝細胞進?NLFRE功能性成分進?各種致病因素致肝損傷護肝作用及抑制肝癌之機制探討,包括抗氧化、抗發炎、促進解毒酵素活化及癌細胞凋亡等,研究架構如附圖一。本研究計劃?能順?達成預定目標,?僅可以有效地充分?用荷?天然的萃取物(NLFE)其功能性成分(NLFRE)?抑制脂肪肝及纖維化及肝癌的形成及機?;證實荷?功能性成分有效的促進體脂肪排泄及抑制合成,抗發炎、抗氧化及促進解毒酵素活化及癌細胞凋亡作用;也證實對多種因素引起肝損傷之保護作用及機轉,進一步產學合作開發為護肝及?低肝癌發生?之保健食品。
    The leaves of Nelumbo nucifera GAERTN (commonly known as lotus) have been utilized for various medicinal purposes in Chinese herbal medicine. Our present studies have mentioned the biological activities of N. nucifera leaves aqueous extracts (NLFE) and N. nucifera leaves flavonoid-rich extracts (NLFRE), including antioxidant and anti-LDL oxidation. In addition, the anti-atherosclerosis effects (supplementary data 1), and the attenuation of hypercholesteremia and lipid accumulation in liver (supplementary data 2) in hamsters by supplementing the N. nucifera leaves extracts into diets were revealed. We also have observed the proliferation and migration of smooth muscle cells and the body fat formation were inhibited effectively after the treatment of the N. nucifera leaves extracts (supplementary data 3 and 4). According to these results, we will further study and develop the NLFE as healthy foods which were applied to delay in fatty liver, protect liver and inhibit liver cancer. Our studies will be divided into several sections. We will study the protection effects of NLFE on different factors-induced liver injury and liver cancer, and the effects and mechanisms of NLFE on eliminating the lipid accumulation in liver fat formation. In first year, we will analyze the liver protective effects of NLFRE on high glucose plus STZ-, APAP-, alcohol-, or LPS-induced liver injury. In second year, we will further study the effects and mechanisms of NLFRE on inhibiting liver fibrosis and liver cancer in animal models. In third year, human hepatocarcinoma HepG2 cells were applied to observe that the protection effects of NLFRE on these different factors-induced liver injury and liver cancer, including antioxidant, anti-inflammation, promoting the activation of detoxifying enzymes, and apoptosis. The research framework is shown in Figure 1. The results of these studies may be used to demonstrate the effects of the N. nucifera leaves extracts on decreasing fatty liver formation, liver fibrosis and liver cancer. We also will confirm the effects and mechanisms of N. nucifera leaves extracts on protecting the different factors-induced liver injury. Further, we will apply the functional components of N. nucifera as healthy foods via the industry-university cooperative research progress.
