    Title: 洛神花青素抑制N-Nitroso-N-Methylurea(NMU)誘發Sprague-Dawley大白鼠急性前骨髓球白血病之研究

    Abstract: 洛神花(Hibiscus sabdariffa Linnaeus)原產非洲等地,本省引種栽培,今以臺東、花蓮等東海岸地區為主要產地。花青素(anthocyanins)廣存於蔬菜、水果等植物中,先前的研究發現花青素具抗氧化活性、降低血脂肪及防癌作用,本研究室也證實分離自洛神花之花青素(Hibiscus anthocyanins,HAs)有抗粥狀動脈硬化及保肝之功能;最近初步的研究顯示,洛神花花青素會促進癌細胞的細胞凋亡,尤其以人類急性前骨髓白血病細胞株(HL-60)最為敏感,因此,本研究藉由動物實驗來探討洛神花花青素預防或延緩癌症之作用。動物模式根據Huggins C.B.等人之研究方法,使用N-Nitroso-N-Methylurea(NMU)誘發Sprague-Dawley大白鼠血癌,由肉眼觀察、劉氏染色、柏氏染色和H&E染色等實驗方法,結果發現洛神花花青素可以抑制NMU誘發血癌芽細胞浸潤大白鼠的肝臟、脾臟及腎臟,型態學上血癌芽細胞的減退以致成熟現象的產生亦清楚可見;此外,為了確立血癌的分型,進一步利用免疫組織化學染色和特殊染色如骨髓過氧化 、CD3、CD20、CD15、CD45、甲型胎兒蛋白和肝醣染色試驗,結果發現為急性前骨髓球白血病。由於洛神花在國內容易栽培,產率高,而洛神花花青素也容易分離,經本研究證實洛神花花青素可延緩或抑制急性前骨髓球白血病的發生,未來希望本研究成果將可應用於發展新藥,然而,對於洛神花花青素抗急性前骨髓球白血病作用的特異性及機制,則有待更進一步的探討。
    Hibiscus sabdariffa Linne (Malvaceae), originally produces places such as Africa. Taiwan introduces a fine variety the cultivation, now take East China Sea shore area and so on the Taitung, Hualian as the main habitat. Anthocyanins exist widely in many vegetables and fruits. As shown in previous studies anthocyanins was found to possess anti-oxidative of activity, inhibiting serum lipids and preventing cancer. Our previous reports demonstrated that anthocyanins extracted from Hibiscus sabdariffa Linne( Hibiscus anthocyanins,HAs) exhibited anti-atherosclerotic effect and liver protection. In the recent research, we explored the effect of HAs on human cancer cells. The result showed that HAs could cause cancer cell apoptosis, especially in HL-60 cells. Therefore, in this study, we evaluated HAs prevent or defer cancer of function by animal experiment. According to the Huggins C.B., we used a model of N-Nitroso-N-Methylurea(NMU)induced Sprague-Dawley rats’ leukemia. The result revealed HAs inhibits leukemia blast infiltration of liver、spleen and kidney induced by NMU in rats, including gross examination, Liu’s stain, Papanicolaou stain and H&E stain. The morphologic finding of maturing elements fell by leukemia blast also it was thus clear. In addition, to determine the classification of leukemia, we studied immunohistochemical stain and special stain as myeloperoxidase、CD3、CD20、CD15、CD45、α-Fetoprotein and Periodic acid-Schiff. Find for the acute promyelocytic leukemia(APL)as a result. Because the Hibiscus sabdariffa Linne educates easily in the home, producing a rate high, and the HAs also extracts easily. According to these findings, we suggest that HAs may be used to defer or repress the occurrence of the acute promyelocytic leukemia. Will hope this research result will be able to be applied in a new medicine of development in the future. However, further investigations into the specificity and mechanism(s) of the function of HAs anti- APL are needed.
