Abstract: | 在本年度計畫中發現長期暴露下發現高劑量下有上皮增生及發炎之現象產生,此外,在腫瘤促進作用之指標亦發現MPO活性有上升之趨勢,ODC蛋白活性也有隨劑量增加而增加,相關的訊息傳遞蛋白,如PI-3K, PKC, MAPK, NF-kB, JNK, P38, c-MYC等有顯著變化,所以我們可以發現長期暴露在高劑量之農藥環境下,存在著腫瘤促進的現象. Trichlorfon a kind of organophosphate insecticide, has been widely used in agriculture as a broad spectrum insecticide based on its action as an inhibitor of acetylcholine esterase. Prochloraz is widely used as an agriculture fungicide. The action od imidazole fungicide is based on the inhibition of cytochrome P-450-dependent step in ergosterol synthesis. In the present project, we hypoesized that chronic exprosure of trichlorfon and prochloraz around skin might induced tumor promotion and absrpotion toxicity, which could cause alteration of serum biochemical in the 2 month, 4 month and 6 month treatment. To investigated the toxicity of trichlorfon and prochloraz in rats, following work will be finished: (1) analysis serum biochemical level; (2) patholgenic examination; (3) marker of tumor promotion assay (inflammation, hyperplasia, ornithaine decarboxylase, myloperoxidase activity, H2O2 and hyperplasia). The results of these studies may be used to demonstrate the organ toxicity and tumor promotion of trichlorfon and prochloraz in rat skin exposure 6 month. |
