戊乙聽去氫襪子甘 [pental acetyl ge poside (AC)jGP] 是從山
棍子 (0缸往enia fruc )萃取出來的一種酵甘(glycosi e) ,再經乙驢化
(acetylation) 而得到的。本實驗室之前的研究指出, (AC)jGP 可以抑
制老鼠體內 品神經膠瘤細胞的生長。本篇實驗進一步利用 C-6
經膠瘤細胞株,處理(AC)jOP 後發現,在處理(AC)jOP 之後,會造
品細胞的死亡、DNA 階梯狀的斷裂及細胞的過期停在 00/G1 翔,
茵而判定細胞(AC)jGP 造成細胞的死亡,是一種細胞凋謝死亡的形
式。探討結果發現, (AC)jGP 會導致 毛細胞內 p53 c-Myc 蛋白
的增加,進而活化下游的 Bax' 並抑制 Bcl-2 的表現,來導致細胞的
獨亡。另外~會經由促進 p53 的表現,進一步促進下游 p21 蛋白表
現並抑制 RB 蛋白,來導致細胞遠期停留在 00/01 期。另外也發現
PKC 轉位活化的情形,但 PKC 下游的一些訊息傳遞蛋白卻不受
(AC)jGP 的影響。而同時給予 TPA H7 處理之後,卻未發現(AC)jGP
達成 C-6 細胞過期停滯的情形有受到抑制。盟此推斷 PKC 並沒有參
(AC)jGP 抑制細胞過期的路徑中。
Penta-acetyl geniposide, (AC)sGP, generated by acetylation of
a glycoside isolated om extracts of Gardenia uctus. In previous
study , we reported lat (AC)sGP could inhib扭曲 grow由組

The (AC)sGP induced cell death was examined C-6 glioma cells.
Treatment of glioma cells with the (AC)sGP caused formation of
intemucleosomal DNA ladder cell cyc1e arrest at GO/G 1 phase. These
phenomenon 拉拉icated 出獄 (AC)sGP-induced cell death appeared to be
mediated by apoptosis. In addition, the results also showed at p53
protein was accumulated by treated with (AC)sGP and cause apoptosis
upregul ion of Bax and downregulation of Bc1-2. On the other hand,
(AC)sGP independent p53 expression could also induce p21 expression
while RB expression was inhìbited and cause cell Test at GO/G 1 ph師In addition, PKC could also be activated this pathway, but lere lS no
effect PKC downstream protein expression. Treating cell with TPA or
nd扭惡也at TPA or H7 can't inhibit the effect of cel1 cyc1e arrest by
(AC)sGP.So we suggest latPKC not involved (AC)sGP hibiting
pathway of cell arrest.
