    Title: 台灣本土五葉松活化免疫與抑癌成份之評估研究-1
    Evaluate the Active Compounds Within the Extracts of Taiwan Local Pinus Morrisonicola Hayata Effective on Immunity and Tumor Suppression-1

    Abstract: 數千年前神農氏選出「長生十傑」:『日、山、水、石、松、雲、木、草、鶴、龜』,其 中「松」列為十傑之一;「神農本草經」描述能維持生命,有助於長壽者視為「上藥」, 「上藥」之首為最常被提到的天然植物藥材:「松」類植物。宋朝「重修政和經史證類 備用本草」一書中,曾有如下之記載「松,可生毛髮、安內臟、癒空腹,長生」;明朝 李時珍著作漢方聖經之稱的「本草綱目」中記載:「松葉名松毛。苦、溫、無毒。能生 髮、安五臟、解飢、延天年」,述說著松葉的藥效能夠治療腫疱(腫瘤與囊疱)、促進毛 髮的再生、強健內臟(肝、腎、脾、心、肺五臟)、充飢及延年益壽。中藥大辭典中記 載,松樹藥材其功用具備袪風濕痹痛、排膿及拔毒功能。然而,臺灣本土五葉松對人體 益處在台灣卻鮮少重視研究。因此,本計畫將取材臺灣本土松屬天然植物,探究五葉松 防癌抑癌的生物生理功能,尤其著重在免疫與癌症預防研究。將進行分析研究臺灣唯有、 本土特殊植物五葉松(白松)萃取物,尋找具備增強免疫調節功能、阻斷癌細胞啟始、 促進及延續進行的天然成份,與抑制腫瘤孳生的優質天然物。免疫力調節評估測試包含 細胞增殖實驗、細胞激素的分析、自然殺手細胞活性、吞噬細胞活性、抗體分泌實驗、 特異性細胞增殖、抗體產生及細胞激素的製造能力;防癌分析包含天然物對抗促進腫瘤 生成因子、偵測細胞內自由基之清除能力及藥物抗氧化力、抗發炎反應、調節藥物代謝 作用能力、單細胞傷害電泳分析;而抑癌研究包含腫瘤生長抑制、腫瘤轉移抑制及新血 管生成抑制之檢測。實驗室過去自臺灣本土松屬、麟毛蕨類、野桐、粗糠柴、葎草與地 耳草中分析純化出各類間苯三酚衍生物,並進行研究得知其衍生物具備抑制癌症生長因 子(鳥胺酸去羧化酶)表現及促進癌細胞走向死亡的功能,因此本計畫將進行專注臺灣 本土五葉松天然植物萃取物製備和成份分離與分析比對,利用質譜儀、核磁共振儀和紅 外光譜鑑定化學結構,篩選有效成份,並建立天然物防癌抑癌生物活性標準化檢測標的 與方法,結合天然物結構、酵素動力及酵素立體結構學,進一步提供天然物結構預測資 訊、尋找專一性抑制特殊標的天然物,同時作為國內外天然物修飾合成的參考,以期尋 找出五葉松中蘊含之優質防癌、抑癌天然成份與其衍生物。終極目標促進保健食品與藥 物開發可能性,再經由雛形商品測試開發,達到量產技術開發的技轉理想。
    In the ancient China’s books, [Shen Nong Ben Cao Jing] and [Bencao Gangmu], mention that plants such as pines, woods and grasses are the best source of drugs for lengthen human life and to be the health foods. People have all along been seeking to prolong life. Pinus morrisonicola Hayata is one of original coniferous species in Taiwan. To investigate the bio‐effects of Pinus morrisonicola Hayata, we will carry out the detailed analysis of the reactive components by using HPLC, GC‐MS, LC‐MS, IR and NMR spectroscopy to isolate and analyze the intermediates and the major or minor compounds that could reveal the potential effects in the enhancing immunity and cancer chemoprevention. The extracts of Pinus morrisonicola Hayata will be evaluated the immune function (immune cell proliferation, the expressions of cytokines and chemokines, the activation of NK cells, macrophage phagocytosis, the population of immune cells, B cell function in vitro and in vivo tests), and the potential of cancer chemoprevention studies (the selection of ornithine decarboxxylase inhibitors, the assay of radical scavenging activity by 1,1‐Diphenyl‐2‐picrylhydrazyl, the superoxide anion scavenging assay, the COX‐2 activity and iNOS activity assay, the modulation of drug metabolism by phase I or II enzymes, cell proliferation and cell differentiation assay, single cell gel electrophoresis, the inhibition of DMBA‐induced preneoplastic lesion formation in mouse mammary organ culture, two‐stage carcinogenesis test, tumorigenesis, metastasis and angiogenesis assay). Besides, we will encourage on the active products of extracts from Pinus morrisonicola Hayata effective on the immune modulation and tumor suppression.
