Abstract: | 目前傳統癌症治療方法分為化學治療及放射線治療;化學治療被稱為最有效的癌症治療方式,是利用一種或多種的藥物來合併使用(又稱為合併性化學治療),而其作用是在破壞癌細胞生長,達到治療癌症、控制病情、緩解症狀的目的。但因化學治療除了殺死癌細胞外,也會對正常生長較快速的細胞造成傷害而產生副作用。而放射線治療即是運用較高能量的放射線,約是診斷用的X光之能量的數百或數千倍,來破壞癌細胞,防止癌細胞的生長與分裂,也是目前治療肺癌的主要策略之一。但對於正常組織也是有劑量耐受性的限制,而放射線對於不同癌細胞也有不同的感受性,且放射線對於癌症病人也具有相當程度的副作用;本研究之目的為利用berberine黃連素此中草藥來合併放射線治療,以期能輔助對肺癌細胞及腫瘤的毒殺性,甚至可以使放射線的劑量可以降低,以降低癌症病人的副作用。在動物實驗中,?用裸鼠在皮下種植人類肺癌細胞A549,發現當?鼠給予berberine 合併放射線時會明顯地抑制腫瘤大小,再以Western blot方法分析蛋白表現,探討其作用的機轉,由實驗結果Bcl-2、caspase-3、ATG-5及LC-3的蛋白表現量顯示,使用黃連素合併放射線實驗中在apotosis及autophage並無顯著的相關性。
