    Title: 龍葵水萃取物在雙氯芬酸誘導大白鼠胃黏膜損傷的預防作用

    Abstract:   雙氯芬酸(Diclofenac)是一種非類固醇抗發炎藥物,口服使用下具有引起胃黏膜損傷的副作用。而文獻指出龍葵具有抗發炎、抗氧化的功能,因此我們想要評估龍葵萃取物是否能預防Diclofenac所造成的胃黏膜損傷。實驗的大白鼠預先給予不同劑量的龍葵水萃取物,和預防藥物氫離子幫浦抑制劑Esomeprazole (500 μg/kg)當作保護參考,再餵食Diclofenac (100 mg/kg)以誘導胃黏膜損傷。藉由分析血液中白血球數量和紅血球中麩胱甘?硫轉移?(GST)活性、胃黏膜脂質過氧化物硫代巴比妥酸反應物質(TBARS)和抗氧化物麩胱甘?(GSH)濃度、以及胃部的病理組織切片檢查,來評估龍葵水萃取物對於胃部的保護功能。將實驗組和Diclofenac測試組作比較,發現實驗組紅血球中GST的活性和胃黏膜中GSH的濃度有顯著提升,而且對加入的龍葵水萃取物呈現dose-dependent的提升;而實驗組白血球的數量和胃黏膜中TBARS的濃度亦有顯著降低,同時也會對加入的龍葵水萃取物呈現dose-dependent的下降。病理組織切片檢查發現,使用龍葵萃取物確實可以預防胃黏膜損傷。綜合以上的結果,龍葵水萃取物能夠預防Diclofenac所引起的胃黏膜損傷,或許是受到其抗氧化特性和捕捉活性氧自由基的能力所影響,因此龍葵水萃取物確實可以運用來作為胃部黏膜的保護劑。
    Diclofenac is a kind of non-steroid anti-inflammation drug. Under oral administration, it has the side effect of gastric mucosal damage. Some literature indicates that Solanum nigrum has functions of anti-inflammatory and anti-oxidation. Therefore, we evaluate whether the Solanum nigrum water extract (SNE) can prevent the gastric mucosal damage caused by Diclofenac. Different dosages of Solanum nigrum water extract, and proton pump inhibitor (Esomeprazole, 500 μg/kg) as preventive medicine for the protection reference are given to the Wistar rat for experiment, then oral administration of Diclofenac (100 mg/kg) inducing gastric mucosal damage. By analysis of white blood cell count, activity of antioxidant enzyme glutathione-S-transferase (GST) in red blood cell, as well as the concentration of gastric mucosal lipid peroxide thiobarbituric acid reactive substances (TBARS) and the antioxidant enzyme glutathione (GSH), as well as histological examination of stomach. The result of our study showed that evaluate the protection for stomach provided by Solanum nigrum water extract. Compared with experiment group and Diclofenac test group, a remarkable promotion on activity of GSTs and the concentration of GSH in experiment group is noted. Moreover, it presents dose-dependent to the Solanum nigrum water extract. Obvious reductions on white blood cell count and the concentration of TBARS in gastric mucosa are also found and present dose-dependent to Solanum nigrum water extract. The pathology examination reveals that treatment of Solanum nigrum water extract may prevent the gastric mucosal damage. To sum up, the Solanum nigrum water extract can prevent the gastric mucosal damage caused by Diclofenac. The results maybe due to antioxidation and the ability of free radical scavenging of Solanum nigrum water extract. The Solanum nigrum water extract surely can be utilized as protecting agent of gastric mucosa.
