    Title: 龍葵水萃物抑制乳癌細胞轉移之研究
    Study on Aqueous of Extract Solanum nigram in Inhibiting Migration in Human Breast Cancer Cell

    Abstract: 乳癌在台灣的癌症死亡統計中排名第五位。龍葵,學名Solanum nigrum L.,始載於唐本草,在台灣常為草藥被使用。龍葵水萃物具有抗氧化、解熱鎮痛及抗發炎的效果,保肝作用與抗癌作用。在最近的研究指出,龍葵水萃物具有抑制肝炎細胞的生長及肝硬化,但對於龍葵是否影響乳癌尚未清楚。本篇研究討論龍葵水萃物對於人類乳癌 MDA-MB-231 細胞的毒性測試及 MDA-MB-231 細胞的轉移現象。將 MDA-MB-231 細胞分別加入不同濃度的龍葵 (0.01、0.05、0.1、0.5、1、3、5 mg/ml) ,在 37℃ 下做用 12、24 小時,利用 MTT assay 測定細胞存活率,確認何種濃度的龍葵處理下不會造成 MDA-MB-231 細胞受損。經由實驗結果得知,當細胞處理龍葵反應 12 及 24 小時後,在 1 mg/mL 濃度時並不會影響細胞的存活,因此選用 0.1、0.5、1 mg/ml 作為本研究的濃度。經實驗結果得知,在控制組中,未處理龍葵的 MDA-MB-231 細胞在 24 小時會將缺口癒合,在對照組當中,當處理了不同濃度的龍葵後,隨著龍葵濃度的增加,缺口癒合的能力下降,因此可以看出,龍葵具有抑制細胞移動的能力。為了更進一步的確認傷口癒合實驗中,龍葵抑制乳癌細胞的移動能力,在此利用 transwell assay 來加以驗證。經24小時的培養後,利用 Giemsa 染色,經顯微鏡下的觀察可以看到當處理了不同濃度的龍葵後,隨著龍葵濃度的增加,細胞的數目會減少,因此可以看出,龍葵具有抑制細胞移動的能力。由量化的結果發現,在 0.5 mg/ml 龍葵就已具有抑制細胞移動的現象。
      利用西方點墨法來探討龍葵引起細胞遷移的作用。從實驗結果發現,將 MDA-MB-231 細胞加入不同龍葵的龍葵 (0.1、0.5、1 mg/ml),在 37℃ 下處理24小時後,隨著龍葵濃度的增加會使得 RhoA、Rac1 和 cdc42蛋白表現量降低,而使得 RhoB 蛋白表現量隨著濃度增加而增加。綜合以上的結果顯示龍葵水萃物可以抑制細胞轉移的現象,但對於是否可成為抗癌藥物,需要再做更深入的研究,評估其療效。
    Breast cancer is the 5th position of death causes in Taiwan. The water extract of Solanum nigrum (SNWE) possess the abilities in anti-oxidation, anti-inflammation, antipyretic, and anti-cancer effects. Recent reports reveals that the water extract of Solanum nigrum (SNWE) inhibits the growth of hepatitis cell and cirrhosis. However, the impact of Solanum nigrum on breast cancer cell remains unclear.
    In this study, we discuss the cyto-toxicity of water extract of Solanum nigrum on human breast cancer cell line MDA-MB-231 and the inhibition on MDA-MB-231 cell migration. We performed MTT assay to test the survival of MDA-MB-231 cell under the treatment of different concentration of Solanum nigrum (0.01、0.05、0.1、0.5、1、3、5 mg/ml). We have found that cell can well survive under the concentration of 1 mg/mL after 12 and 24 hours of treatment, so we chose concentration of 0.1、0.5、1 mg/ml to perform following cell migration assay. Solanum nigrum-untreated MDA-MB-231 cell will heal after 24 hours. The healing ability decreases while treated with Solanum nigrum. To further test the migration inhibition of Solanum nigrum on MDA-MB-231 cell, we performed transwell assay and found that Solanum nigrum can inhibit the migration of cancer cell under the concentration of 0.5 mg/ml.
    Western blot reveals that when treated with different concentration of Solanum nigrum (0.1、0.5、1 mg/ml), the protein expression of RhoA, Rac1 and cdc42 in MDA-MB-231 cell decreases while the protein expression of RhoB increases. We conclude that the water extract of Solanum nigrum inhibit cell migration. Whether it can serve as anti-cancer drugs remains further research.
