    Abstract: 肺癌是包括台灣之內世界癌症患者的首要殺手,其死亡人數約佔全世界癌症死亡人數的百分之二十.抽煙與百分之九十之肺癌肺癌形成有關, 約有百分之十的重度吸煙者罹患肺癌.在台灣,女性肺癌約佔總肺癌人數的百分之三十,奇特的是絕大部分的患者事終生不吸煙者.過去,肺癌機制的研究都偏重於與抽煙強烈有關的肺癌,如小細胞肺癌和麟狀表皮細胞肺癌.對於台灣女性罹患率高的肺腺癌形成的原因尚未了解.肺癌的五年存活率不到百分之十五.如果早期診斷除肺癌,其五年的存活率課題高至百分之五十.因此發展早期診斷的方法和研究先進的治療方法,對於提高肺癌的存活率是同等的重要.而這兩項任務都需要對細胞生物學和分子生物學有充分的了解.因此,在分子層次上有系統的尋找參與肺癌致病過程的基因,可能是提升肺癌早期診斷和治療的重要途徑.本實驗是已建立兩個肺腺癌的精簡基因庫,並積極的展開鑑定一些可能與肺腺癌發生有關的致癌基因和抑癌基因.
    Lung cancer has been the leading cause of cancer deaths worldwide, and it account for about 20% of total cancer deaths. The mortality rate of lung cancer has also been the highest in Taiwan. Cigarette smoking is attributed to about 90% of lung cancer. About 10% of the heavy smokers develop lung cancer. In Taiwan, about 30% of lung cancers are female and more than 90% of them have never smoked. About 70% of female lung cancers in Taiwan are adenocarcinoma. The mechanism of developing lung adenocarcinoma in non-smokers is still unknown. The high mortality rate of lung cancer is not due to high incident rate. It is partly due to lack of the early detection and partly resulting from low response to chemotherapy. The average of 5-year survival rate of lung cancer is less than 15%, however the 5-year survival rate of early stage lung cancer can be increased to over 50%. Thus, developments of early detection methods as well as of new treatments are equally important to increase the survival rate of lung cancers. Both tasks require advance knowledge in cell biology and molecular biology of lung cancers. First step toward understanding the molecular biology of lung cancer is to identify genes that are important to the carcinogenic process of lung cancer. To reach the goal, we have established two subtracted lung adenocarcinoma cDNA libraries from a single patient and in the process of identifying clones in the subtracted cDNA libraries. A cDNA microarray will be generated form clones in these subtracted libraries. The cDNA microarray will be used for studying carcinogenic process of lung adenocarcinoma and for screening of patients. This project aims to identify genes that are specifically related to carcinogenic process of lung adenocarcinoma, and genes that are responsible for resistant to chemotherapy. We will prioritize and characterize few genes for further functional studies.
