    Title: 牛樟芝子實體之創新固態栽培技術的研發與活性成分之研究

    Abstract: 研究結果顯示,樟芝發酵液抑制癌細胞生長功效較佳;由DNA斷裂分析得知樟芝水萃物與甲醇萃取物則可較有效地誘發癌細胞之細胞自戕作用,而樟芝發酵液與樟芝水萃物則可有效抑制癌細胞在半固態培養基中形成細胞株群落的活性。提升免疫反應方面,樟芝發酵液、人工樟芝水萃物、人工樟芝甲醇萃取物與野生樟芝甲醇萃取物對癌細胞都有影響,惟野生樟芝水萃物對癌細胞的影響無顯著差異。本實驗室之研究成果不但已建立牛樟芝的最佳人工栽培條件與技術,並已證實在實驗室利用人工栽培之牛樟芝具有抗腫瘤細胞生長與誘發細胞自戕作用的活性。
    Antrodia cinnamomea is a unique brown-rot fungus species which only parasitizes in the decayed heart wood of stout camphor (Cinnamomum kanehirae) in Taiwan. The fruiting body of A. cinnamomea contains abundant polysaccharrides and terpenes that are proved to have effective anti-viral function and anti-tumor activity. The price of A. cinnamomea therefore rocketed in the market and it has become the target of illegal harvesting. Consequently, C. kanehirae has become an endangered species in Taiwan due to the illegal felling. The present proposal is a 2-year project that is divided into 2 stages: the first-year project, mainly conducted by professionals from the Department of Forest Products Science and Department of Forestry and Natural Resources in National Chia-Yi University, is aimed to develop novel solid culture techniques for cultivating A. cinnamomea. The second-year project is executed by professionals in School of Medical Laboratory and Biotechnology, Chung Shan Medical University, and Division of Basic Medicine of Hung Kuang University. The major theme is to identify the effective and active anti-tumor and immune-promoting component(s) of A. cinnamomea which is cultured and provided in Lab. by the first year project. The active components or the ratio of secondary metabolites of A. cinnamomea derived from liquid or solid culture will be further compared in the second stage of the project for the establishment of solid culture techniques that can nurture A. cinnamomea with high content of active components in large scale. The implementation of thi project can not only uncover the mechanism of anti-tumor and immune-promoting components from A. cinnamomea, but also establish effective solid culture techniques that can culture A. cinnamomea with high content of active components and secondary metabolites in large scale. By establishing the effective solid culture techniques to produce A. cinnamomea in large scale, the price of A. cinnamomea can not only be decreased, the crisis in which C. kanehirae extinction resulted from illegally harvesting A. cinnamomea as well as the destroy of forest ecology in Taiwan can also be saved and solved.
