    Title: AH26與AHplus根管充填劑之溶出物分析及致突出病變性研究

    Abstract: 牙科用根管充填劑AH 26和AH plus為以環氧樹脂為基底的充填材料。過去研究證明AH 26於混合後會釋出Formaldehyde,造成細胞毒性與基因毒性。本實驗室對於AH 26與AH plus的研究發現,以MTT assay作用於人類口腔上皮癌細胞株,結果均具有細胞毒性。另外,AH 26經Ames test,Nick translation assay和Sos chromatography assay均證明據有基因毒性。由過去本實驗室研究AH 26之釋出物,於一年後仍然具有細胞毒性,我們懷疑是否還有其它物質會造成細胞傷害。根據廠商宣稱AH plus不會釋出Formaldehyde,那究竟是何種物質造成細胞毒性?因此本實驗以HPLC分析,LC-MS分析來研究AH 26和AH plus的釋出物。結果發現AH 26會釋出Formaldehyde,而AH Plus則不會釋出;另外,AH 26和AH plus之釋出物作用於OC2則具有不同程度之基因毒性出現。
    AH 26 and AH plus are epoxy resin type root canal filling materials. The previous studies demonstrated that the AH 26 may cause the cytotoxicity and mutagenicity by releasing the formaldehyde. AH plus is a modified root filling material. According to the manufactor's describe that AH plus will not release the formaldehyde. From our past studies on the cytotoxicity of the AH 26 and AH plus. The result showed that both AH 26 and AH plus were toxic to OC2 cell line by MTT assay. We doubt that except the formaldehyde there might be others components release from the AH 26. Also, what component released from the AH plus to cause the toxicity is interesting to us. By using the HPLC analysis, the finding showed that the AH 26 releasing the formaldehyde can be detected, but not the AH plus. The actual component of the AH plus releasing material need to analysis again. The genotoxicity of the AH 26 and AH plus showed degrees of genotoxicity by comet analysis.
