    Title: CRBP1蛋白在人類癌症中的表現

    Abstract: 大腸直腸癌是台灣第二常見的惡性腫瘤,也是位居第三名的癌症死亡病因。大部份的癌症死因通常是惡性的進程及轉移所造成的。大腸直腸癌的致癌因子雖然眾所皆知,但其基因的變化及轉移的機轉,目前還不是很清楚。而大約有百分之七十五至八十的病人在疾病被診斷出來時,病灶仍侷限在腸壁或是周遭淋巴結,並未轉移。視黃醇接合蛋白(celluar retinol binding protein1, CRBP1) 是一個視黃醇的胞外運送蛋白。它在很多的組織中都會表現,但是在各種組織中其濃度變化是非常大的。有paper指出,CRBP1在人類乳癌中,會有較低的表現,且會抑制乳癌細胞株的增殖情形,故有人推測CRBP1蛋白可能有雙重功能,其一是與視黃醇酸的代謝有關,其二是與腫瘤的抑制具有相關性。本篇研究使用組織免疫化學染色對大腸直腸癌的病人進行CRBP1染色,結果顯示大部份的大腸直腸癌病人為陰性表現,少部份呈現陽性表現。研究中亦統計了病人的年齡、性別、癌症分期、及TMN分類,結果發現,在癌症分期中,將分化良好腺癌與中度分化腺癌為一群,分化不良的腺癌為一群,互相做比較,其統計上是有意義的(P<0.05)。總而言之,本篇利用IHC提供了CRBP1在大腸直腸癌中的表現情況,推測或許CRBP1在大腸直腸癌的發展中,扮演著重要角色。
    Colorectal Cancer(CRC) is the second most common malignancy in Taiwan and the third most frequent cause of cancer-related death. Most of cancer death is due to malignant progression and metastasis. The tumorogenesis of CRC is well known, but the genetic alternations and mechanisms of metastasis of CRC are largely unknown. Approximately 75%-80% of patients are diagnosed with disease limited to the bowel wall or surrounding lymph nodes. Cellular retinol binding protein 1 (CRBP1) is the cytosolic carrier for retinol. It is expressed in many tissues, but the concentrations vary considerably. CRBP1 is under-expressed in 24% of human breast carcinomas, and has also been shown to suppress proliferation and anchorage-independent survival of a breast cancer cell line , suggests that this protein may have a dual function in retinoic acid metabolism and tumor suppression. We used the immunochemistry stain (IHC) dyeing looked the CRBP1 protein in the test sample of the colonrectal cancer patient. The result shows that the majority of colonrectal cancer all had not displayed, only few colonrectal cancer were positive performance. We statistic the patients age, gender, grading, TMN classification, and the cancer classification between well-differentiation with moderate-differentiation colonrectal carcinoma and pooly-differentiation carcinoma are related (P<0.05).Taken together, our results provided the expression of CRBP1 in colonrectal cancer. It suggested that this gene may play an important role in colonretcal cancer development.
