    摘要: 三氧礦化合物為一生物相容性之根尖充填材料,一種已開發之新的鈣矽類化合物,其物理性質和化學性質與傳統之三氧礦化合物相近。本研究目的為研究此鈣矽類化合物與傳統之三氧礦化合物對於細胞之生物相容性。研究方法:鈣矽類化合物與傳統之三氧礦化合物經混合配製後,分別與老鼠纖維細胞株(mouse fibroblast L929)一起培養一天、三天、七天和十五天後,以粒線體呈色反應分析(mitochondrial colorimetric assay)其細胞之存活率,並以電子顯微鏡觀察細胞與材料之附著形態變化。西方墨點分析則來評估老鼠纖維細胞株與材料作用後其細胞內訊息蛋白ERK和磷酸化ERK之變化。結果分析則以ANOVA分析不同材料下之差異,結果顯示老鼠纖維細胞株於矽類化合物與傳統之三氧礦化合物之存活率均高於控制組。電子顯微鏡觀察下,細胞可良好生長附著於材料上,具細胞相容性。細胞內訊息蛋白ERK和磷酸化ERK的表現則同樣明顯出現於鈣矽類化合物與傳統之三氧礦化合物之細胞處理上。結論:鈣矽類化合物與三氧礦化合物一樣,對於老鼠纖維細胞株具有相同之生物相容性。
    Mineral trioxide aggregate (MTA) is a biocompatible endodontic retrofilling material. A recently developed calcium silicate (CS) cement has also shown similar physical and chemical properties as MTA. The purpose of the present study was to compare the biocompatibility of MTA and CS with mouse fibroblast (L929) cell line. MTA cements and 4 CS cement group made of different components were cultured with L929 for 1, 3, 7 and 15 days. L929 cell survival rate was measured by mitochondria colorimetric assay (MTT), the morphology of the cells growing on the cements were observed by scan electromicroscope, and the expression ERk and pERk L929 cell measured by Western blot analysis. One way analysis of variance (ANOVA) was used to determine statistical difference. A p<0.05 was considered significant. Both MTA and CS cement were found to be equally biocompatible with L929 cells. L929 cell attached well to both MTA and CS cements. The L929 expression of ERk and pERk was the same for both groups. We conclude that CS and MTA share the same biologic properties and are equally biocompatible with L929 cells.
