    Abstract: 動脈粥狀硬化是引起全世界冠狀動脈心臟病(CAD)死亡的主要原因,包括台灣。心血管疾病是由多種因素,包括遺傳因素和環境因素之間的相互作用所引起。主要環境因素包括高血壓、糖尿病、高脂血症、肥胖、吸菸以及飲酒…等。然而這些因素不能完全地預測心血管疾病發生,因此遺傳基因可能有助於發現心血管疾病的潛在發病機制。越來越多的證據表明,炎症在動脈粥狀硬化的發生和發展中起著重要作用,動脈粥狀硬化被認為是一種進行性炎症疾病。當動脈內皮受損時,會引起局部性的發炎反應,IL-1是主要的促炎細胞激素,其中IL-1β在動脈粥狀硬化和CAD中起重要作用。因此本篇研究的目的主要是想了解台灣人群中,IL-1β-511C/T基因多型性與動脈粥狀硬化風險之間的關聯性。本次研究使用病例-控制實驗的方法,參與研究的樣本數目共有199人,其中100人為動脈粥狀硬化病例組,另99人為無冠狀動脈硬化症的正常人對照組。我們利用PCR-RFLP的方法來偵測IL-1β-511 C/T基因多型性。結果表明IL-1β-511C / T基因型分佈和等位基因頻率與動脈粥狀硬化風險之間無顯著相關性。在動脈粥狀硬化的患者中,攜帶IL-1β-511 CT+TT基因型得到糖尿病風險是攜帶CC基因型者的3.077倍,ORs (95%CI)為3.077 (1.020-9.281)。因此,我們推論在台灣人群中,IL-1β-511 C/T基因多型性可能與動脈粥狀硬化風險增加無顯著相關性。我們的研究發現攜帶IL-1β-511 CT +TT基因型的動脈粥狀硬化患者與糖尿病風險增加有顯著相關性。
    Atherosclerosis is the leading cause of coronary heart disease (CAD) death worldwide, including Taiwan. Cardiovascular disease is caused by a combination of factors, including genetic and environmental factors. The main environmental factors include hypertension, diabetes mellitus, hyperlipidemia, obesity, smoking, and drinking. However, these factors do not fully predict the occurrence of cardiovascular disease, so genetic factors may help to discover the underlying pathogenesis of cardiovascular disease. There is increasing evidence that inflammation plays an important role in the development and progression of atherosclerosis, which is considered a progressive inflammatory disease. When the arterial endothelium is damaged, it causes a local inflammatory response. IL-1 is the main pro-inflammatory cytokine, and IL-1β plays an important role in atherosclerosis and CAD. Therefore, the purpose of this study is to understand the association between the polymorphism of IL-1β-511C/T gene and the risk of atherosclerosis in the Taiwanese ethnic group. In this study, a case-control experiment was used. The number of samples involved in the study was 199, of which 100 were atherosclerotic cases and 99 were normal controls without coronary atherosclerosis. We used PCR-RFLP to detect IL-1β-511 C/T gene polymorphism. The results showed that there was no significant correlation between IL-1β-511C / T genotype distribution and allele frequency with the risk of atherosclerosis.
    In patients with atherosclerosis, the risk of diabetes with the IL-1β-511 CT+TT genotype was 3.077 times higher than that of the CC genotype, and ORs (95% CI) was 3.077 (1.020-9.281). Therefore, we conclude that the IL-1β-511 C/T gene polymorphism may not be significantly associated with an increased risk of atherosclerosis in the Taiwanese population. Our study found that patients with atherosclerosis carrying the IL-1β-511 CT+TT genotype were significantly associated with an increased risk of diabetes.
