    Title: 以液相層析串聯質譜儀分析紙片中濫用藥物
    Determination drugs of abuse in paper using High Performance Liquid Chromatography Tandem Mass Spectrometry

    Abstract: 尿液檢測是目前全世界濫用藥物檢驗最主要的方式,但是尿液檢體的運送與長期儲存常有其不便之處。若尿液檢體也能像新生兒遺傳疾病之採血片一樣保存於紙片中,檢體在配送以及保存就會方便許多。本研究針對國內常見濫用藥物,包含安非他命類、搖頭丸、愷他命與海洛因,發展使用紙片來儲存尿液檢體。研究首先建立液相串聯層析質譜儀(LC-MS/MS)檢驗方法,隨後針對乾紙片中各濫用藥物之萃取效率評估,找出最佳材質的濾紙與萃取條件,最後進行儲存溫度之評估。結果發現以甲醇浸泡有80?90%回收率。各藥物的方法定量極限為安非他命:3 ng/mL,甲基安非他命:3 ng/mL,MDA:4 ng/mL,MDMA:4 ng/mL,嗎啡:10 ng/mL,可待因:10 ng/mL,去甲基愷他命:2 ng/mL,愷他命:2 ng/mL。檢量線之線性範圍在25-1000 ng/mL,各藥物線性回歸係數(r2)皆可大於0.995,而長期儲存溫度之評估發現放置於4 ℃之環境下,藥物即不易降解。最後將乾紙片之方法應用於在快速篩檢試紙的另類分析,針對整片或樣品進樣部位進行萃取,隨後進行多重藥物之LC-MS/MS之微量分析,即可獲得嫌疑犯施用之濫用藥物更多資訊。Urine is the most common specimen for drugs of abuse testing specimen, but the shortcoming is needed to refrigerate while transporting and storing. It will also cause the cost increasing while the urine samples from abroad. As dried blood spots generally used in neonate's genetic disease screening, the easily transport dried urine spots (DUS) with the advantage apt to store and transport, but does not have relevant assessment yet in the drug of abuse at present. In this study, it will be developed DUS LC-MS/MS for drug testing. The study focus on the common drugs of abuse in Taiwan, including Mor, Cod, MA, Amp, MDMA, MDA, K, and NK. At first, the ability of the urine, assessing at degree, dry speed of quantitative filter paper. Secondly, the recovery was assessed the incubation solution. The method showed acceptable precisions and accuracy of the expected value, respectively. The limit of quantification (LOQ) values which were sufficiently low to allow whole analytes to be detected with linear range from 25 to 1000 ng/mL. The stability of dried urine spots in different stored temperature was evaluated. Finally, the DUS LC-MS/MS method was applied to Lateral flow assay for reexamined the multiple drugs of abuse simultaneously in the sample pad.
