    Title: 表兒茶素沒食子酸抑制A549人類肺癌細胞之侵襲與轉移活性之研究

    Abstract: 癌症佔據國人十大死因之首已久,肺癌雖不為發生率最高之癌症,但卻是致死率最高的癌症。肺癌的轉移被視為預後的關鍵指標,一旦發生腫瘤轉移,會提高致死率。腫瘤轉移與腫瘤細胞的移動能力與侵襲能力相關,當腫瘤細胞的移動能力與侵襲能力被抑制後,腫瘤轉移也能同時被抑制。綠茶為國人最常見的飲品之一,綠茶中豐富的多酚類物質使綠茶具有抗氧化、抗癌、抗血脂的功能,其中兒茶素是含量最高的多酚物質。主要4種兒茶素有(-)-epicatechin (EC)、(-)-epicatechin gallate (ECG)、(-) - epigallocatechin (EGC)及(-) –epigalloncatechin -3-gallate (EGCG)。其中EGCG為綠茶中含量最高的兒茶素,已有許多研究證實,ECGC能抑制腫瘤細胞的轉移。ECG雖不是含量最高的兒茶素,少數研究指出,ECG可降低心血管疾病,然而對於ECG在抑制肺癌轉移的影響尚未被深入探討。本篇研究即以人類肺癌細胞株A549為實驗對象,探討ECG對於人類肺癌細胞株A549的侵襲力與移動力之影響。實驗結果發現,經ECG處理人類肺癌細胞株A549後,腫瘤細胞的移動能力與侵襲能力均被抑制。此外,更進一步利用實驗證實,ECG能抑制細胞中的MMP-2 (matrix metalloproteinases -2) 及u-PA (urokinase-type plasminogen activator)的表現,使細胞外基質被降解的活動受到抑制,影響了細胞的侵襲能力與移動能力,進而抑制腫瘤細胞的轉移。In Taiwan, cancer is the first of the top ten causes of death for a long time. Although lung cancer is not the highest incidence of cancer, but it is the highest mortality rate of cancer. Metastasis is a key of tumor prognosis. When the tumor metastasis, the mortality will increase. And tumor metastasis is affected by cell migration and invasion. When cell migration and invasion is inhibited, tumor metastasis is inhibited.Green tea, one of the popular beverage, contain rich polyphenols, so that with antioxidant, anti-cancer, anti-lipid function. Catechins are the highest content of polyphenols. The four major catechins are (-)-epicatechin (EC), (-)-epicatechin gallate (EGC), (-) - epigallocatechin (EGC) and (-) –epigalloncatechin -3-gallate (EGCG). EGCG is the most abundant polyphenol in green tea, also the most widely studied compound in green tea. Previous studies show that EGCG could inhibit the invasion of human cancer cell. Although ECG is not the most abundant polyphenol in green tea, there are few study demonstrate ECG can anti-carcinogenic. However, the role of ECG in lung cancer metastasis is still unclear. Thus, we aimed to investigate the mechanism of ECG on the activity of cell migration and invasion in human lung cancer A549 cell lines. Our results revel that the inhibitory effects of ECG on cell migration and invasion in A549 cell lines. In addition, we demonstrated ECG inhibit cell migration and invasion via suppressing the expression of matrix metalloproteinases(MMP-2) and urokinase-type plasminogen activator(u-PA).
