    Title: 苧麻根萃取物抑制癌細胞生長,發炎反應降低及癌細胞抗藥性

    Abstract: 苧麻(Boehmeria nivea L. Gaud)也被稱為中國草,多年生草本植物類,屬於蕁麻科。內富含蛋白質、多酚類、黃酮類等。苧麻具有多種治療療效(包括保肝作用)的悠久歷史。然而,沒有文章描述口腔癌細胞研究在B.nivea的抗發炎作用。苧麻對癌症的反應機制也是了解甚少。證據指出苧麻根能夠有效地降低和細胞之生長相關的訊息蛋白質的表現量,如β -catenin,以及減少和細胞生長有關的蛋白質的功能活性,如 phospho-AKT。證據指出 COX-2 和多種疾病的發生有關,比如腫瘤生成和愛滋海默症(Alzheimers disease, AD) 。研究方法及資料:在這項研究中,我們使用口腔表皮癌細胞 (KB cells)、大腸直腸癌(HT-29)、肝癌(Hep3B),及骨癌(U2OS)三個細胞株,分別細胞培養加入藥物。我們製備了苧麻50%丙酮和50%水提取物,通過測量癌細胞數軟瓊脂培養測定(Soft agar Assay),評估其細胞活性,其在(0、0.02、0.2、2mg/ml苧麻 )刺激下。通過用蛋白質電泳技術方法,利收取蛋白質來測定各個標記蛋白的蛋白質分析,評估其抗炎活性、抗藥性、細胞通透性轉移預後治療。研究結果:這些分析指出苧麻根溶解液可以抑制癌細胞生長和癌細胞標記物的發展, 我們亦證實了利用癌細胞在軟瓊脂集落培養測定 (Soft agar Assay)實驗中給予苧麻根溶解液而有效的抑制癌細胞生長和癌細胞的數目。研究顯示發現苧麻根溶解液可抑制癌細胞P-醣蛋白 (P-glycoprotein, P-gp)的表現,此醣蛋白位於細胞膜上調節抗藥性的機轉和影響藥物的通透性相關,所以苧麻根溶解液可以降低癌細胞的抗藥能力。結論與建議:我們發現苧麻根溶解液能夠減緩發炎反應,抑制癌細胞生長,並且能降低癌細胞的抗藥性。所以苧麻根溶解液用來預防治療發炎及癌症是一種新選擇,而繼續提供未來探討苧麻天然物的研究。Objective:Ramie (Boehmeria nivea L. Gaud) is also known as Chinese grass, perennial herbaceous plants, belonging to the nettle family. Rich in protein, polyphenols, flavonoids and so on. Evidence suggests that COX-2 is associated with the occurrence of multiple diseases, such as tumorigenesis and Alzheimers disease (AD). Methods and Materials: In this study, we used oral cell carcinoma cells (KB cells), colorectal cancer (HT-29), liver cancer (Hep3B), and bone cancer (U2OS) three cell lines, respectively, cell culture Add drugs. We prepared 50% acetone and 50% water extract of ramie, and evaluated the cell viability by measuring the number of cancer cells (Soft agar Assay), which was stimulated with (0,0.02,0.2,2 mg / ml ramie) The Protein protein analysis was used to determine the protein analysis of each marker protein by protein electrophoresis, and its anti-inflammatory activity, drug resistance and cell permeability metastasis prognosis were evaluated.Results: These analyzes indicate that ramie root extract can inhibit the growth of cancer cells and the development of cancer cell markers. We have also confirmed that the use of cancer cells in the soft agar assay assay (Soft agar Assay) experiment to give ramie root extract and effective The inhibition of cancer cell growth and the number of cancer cells. Studies have shown that ramie root extract can inhibit the expression of P-glycoprotein , which is located on the cell membrane to regulate drug resistance and affect the permeability of the drug, so ramie Root extract can reduce the ability of cancer cells to resist.
    Conclusion and Suggestion: We found that ramie root extract can slow down the inflammatory response, inhibit the growth of cancer cells, and reduce the resistance of cancer cells. So the ramie root extract is used to prevent the treatment of inflammation and cancer is a new choice, and continue to provide the future study of ramie natural research
