    Title: 以餵食油炸沙拉油之大鼠為模式探討荖葉萃取物對於主動脈血管組織的影響

    Abstract: 熱氧化的油、特別是反覆使用過的油炸油,與一些疾病是有相關性的,包括高血壓、脂肪肝、心血管疾病等。而對於荖葉的相關研究中,已經證實了具有抗氧化、抗癌、抗自由基、抗發炎與保護肝臟的效用。如能將荖葉萃取物融入於我們的生活飲食,對於身體健康應當有所幫助。因此本研究的目的是以餵食油炸過的沙拉油的大鼠為模式,探討荖葉萃取物對於主動脈血管組織的影響。Sprague- Dawley雄性大鼠隨機分為五組: 正常飼料組(Control)、高油脂沙拉油組(USO)、高油脂油炸沙拉油組(FSO)、高油脂油炸低劑量荖葉(FSOL, FSO+Piper betle [50 mg/kg])、高油脂油炸高劑量荖葉(FSOH, FSO+Piper betle [100 mg/kg])。餵食10週後將大鼠犧牲,取主動脈弓組織進行包埋切片並做免疫組織化學染色分析Caspase-3,LC-3和Ki-67三種蛋白的表現。實驗結果顯示,荖葉萃取物對於主動脈血管組織方面和Caspase-3,LC-3與Ki-67的表現未能觀察到統計上明顯的影響,但仍有潛在的效果存在,可能是因為個體之間的差異性過大,而且樣本數不足所造成。故本實驗雖未能得到明確結果,但可作為未來研究的一個參考基礎,經過修正實驗細節,期望往後能有更明確的結論。
    Thermal oxidative oil, especially repeatedly used frying oil, has been implicated in several pathological processes including hypertension, fatty liver, cardiovascular disease. The studies of Piper betel L. (PBL) have confirmed its antioxidant, anticancer, anti-free radicals, anti-inflammatory, and hepatoprotective effects. Therefore, the integration of PBL extracts into daily diet would be beneficial to our life. The aim of the present study is to investigate the effects of PBL extracts on the aortic vascular tissue using a rat model feeding with thermally oxidized soybean oil. Male Sprague-Dawley rats were divided into five groups:basal diet (control), unused soybean oil diet (USO), soybean oil repeatedly used for deep frying diet (FSO), FSO diet and Piper betel (50 mg/kg) (FSOL), FSO diet and Piper betel (100 mg/kg) (FSOH). After 10 weeks, the rats were sacrificed; the aortic arch tissues were removed and embedded for further analysis. Tissue sections were analyzed by immunohistochemistry for the expressions of three proteins, Caspase- 3, LC-3 and Ki-67. The experiment results showed that PBL extracts affected potentially the expression of Caspase- 3, LC-3 and Ki-67 in aortic vascular tissue although it was not statistical significant. It might be due to large iindividual differences and limited number of samples. Although this experiment failed to obtain conclusive results, it can be used as a reference basis for future studies. We are in hope of acquiring a more explicit conclusion in the future after the modification of some experimental detail.
