    Title: 從牙周病到胰臟癌---研究致病機制與預防(I)
    From Periodontitis to Pancreatic Cancer---Study on the Pathogenesis and Prophylaxis (I)

    Abstract: 本新開拓之轉譯研究主要目的在研究牙周病菌之牙龈卟啉單胞菌 (Prophylomonus gingivalis; P. gingivalis) 在胰臟癌的發展所扮演的角色與預防對策。本計畫中,將自全民健康保險研究資料庫比對胰臟癌患者與其牙周病嚴重程度之相關性,並自胰臟癌患者採集牙菌斑以分離牙龈卟啉單胞菌株,並由共同合作之財團法人食品工業發展研究所進行16S rRNA序列比對,檢討胰臟癌患者所帶此菌基因型與一般人是否有差異。牙龈卟啉單胞菌引發胰臟炎到胰臟癌的致病機制乃運用KrasG12DIpf1-Cre 基因轉殖小鼠的胰臟癌動物模式進行。為了進一步了解排除感染此菌後是否減緩胰臟癌的進程,我們將運用死菌作為疫苗,先行引起小鼠產生免疫力以做為預防。另外,也透過組織病理學的檢驗,如H&E染色、發炎及癌症指標之免疫組織染色,確認胰臟癌的嚴重程度,並由本校病理醫師協助鑑定與確認。可預期本計畫由基礎研究所得之成果將可快速轉譯用於臨床醫學之實證應用。
    This frontier translational research aims to investigate the roles of periodontal pathogen Prophylomonus gingivalis (P. gingivalis) in the development and prophylaxis of pancreatic cancer (PC). In this proposal, the severity of periodontitis of patients with PC will be analyzed using the data gathered from National Health Insurance Research Database, and the genotyping of P. gingivalis strains from PC patients’ dental plaques will be detected with molecular approaches using 16S rRNA sequences in the co-laboratory of Food Industry Research and Development Institute. The pathogenesis of P. gingivalis from pancreatitis to PC will be explored with KrasG12DIpf1-Cre transgenic disease animal model. To generating protective immunity, a dead P. gingivalis vaccine treatment will be given to mice in order to gain a better understanding of whether eliminate the infection would reduce tumor promoting effect of P. gingivalis. The severity of PC will be assayed with histopathologic examinations including H&E stain, immunohistostaining of inflammatory and cancer markers, and the data will be identified by pathologist in our institution. It is expected that the findings of this research will be quickly translated into practical applications in clinical medicine.
